You'd have to mount it as a filesystem. Look up JungleDisk or Infinidisk for S3.
We can't help you implement this -- it would take a great deal of ingenuity and smarts. But I believe it is possible, if you can get S3 mounted as a filesystem, to use Zenphoto with it. No promises on performance either, as it would be very slow.
